It does work, but only on certain older, crappier cars, and a lot of foreign cars. However, the inside of American car doors is not air-tight. It doesn't matter how much air pressure you generate, because it's just going to disapate throughout the rest of the door.

I don't usually comment on the Fake/Non Fake arguments, but this was tried on Mythbusters and it doesn't work, there is someone out of camera shot who presses a button on their car keys, which remotely unlocks the car.
